From vial to syringe: the reconstitution math this calculator runs

Every number this tool returns comes from two inputs you supply: the milligrams of lyophilized sermorelin in the vial and the milliliters of bacteriostatic water you add to dissolve it. Those two figures set the concentration, and concentration is what converts a target dose in micrograms into a mark on the syringe barrel.

Work the default scenario by hand to see the logic. A common sermorelin vial holds 3mg, which is 3,000mcg. Reconstitute it with 3mL of bacteriostatic water and the concentration is 3,000mcg divided by 3mL, or 1,000mcg per mL. On a U-100 insulin syringe, 1mL is marked as 100 units, so each single unit holds 0.01mL, which at this concentration carries 10mcg. A 300mcg dose is therefore 300 divided by 10, or 30 units, drawn to the 30 mark as 0.30mL.

Change one input and the mark moves. Dissolve the same 3mg vial in 1.5mL instead and concentration doubles to 2,000mcg per mL, so each unit now carries 20mcg and the identical 300mcg dose sits at the 15 mark. The dose reaching your body is unchanged. Only the geometry on the barrel changed. That is why the water-volume field is not cosmetic.

Why the unit mark has to be exact

A U-100 unit is a measure of volume, 0.01mL, not a mass. The calculator does the mass-to-volume translation so you read a mark rather than estimate. Precision matters because sermorelin is dosed in the low hundreds of micrograms, where a small barrel error is a large percentage error: at 1,000mcg per mL, a three-unit misread is 30mcg, a full ten percent of a 300mcg dose.

The most common measurement mistakes:

- **Confusing micrograms and milligrams.** 300mcg is 0.3mg. Reading a milligram label figure as the dose is a 1,000-fold error. - **Reusing someone else's unit number.** Their mark only applies if their vial size and water volume match yours exactly. - **Reading units as micrograms.** The 30 mark is 30 units of volume, not 30mcg. - **Ignoring syringe dead space and trapped air bubbles**, which displace volume and shave the delivered dose.

What a 10 to 20 minute half-life dictates about timing

Sermorelin is the first 29 amino acids of growth hormone releasing hormone, and it clears the bloodstream in roughly 10 to 20 minutes, degraded rapidly at its N-terminus by the DPP-4 enzyme. That short half-life is the reason the calculator's default schedule is a single subcutaneous dose at bedtime rather than a steady drip.

The pharmacology drives the schedule in three ways. First, a brief pulse mimics the body's own episodic GHRH signaling, and pulsatile exposure is what keeps pituitary somatotroph cells responsive, whereas continuous exposure tends to desensitize them.
